LuCaS+ is a multicenter single-cell and single-nucleus RNA-sequencing atlas of non-small-cell lung cancer (NSCLC) that captures the cellular and molecular diversity of this disease across clinical stages and histological subtypes. It comprises more than 2.5 million nuclei from 454 patients with extensive clinical annotation and long-term follow-up, generated from formalin-fixed, paraffin-embedded (FFPE) samples at three German university hospitals. The resource is divided into a discovery cohort (Berlin & Cologne, n = 364) and an independent validation cohort (Munich, n = 90), which have been sampled and analyzed seperately, enabling both biological discovery and external confirmation of results.
By integrating high-resolution single-cell transcriptomic data with detailed clinical and pathological information, LuCaS+ reveals tumor subpopulations with distinct metastatic potential, microenvironmental states associated with outcome, and molecular programs linking oncogenic signaling, epithelial–mesenchymal transition, angiogenesis, and immune evasion.
