What is LuCaS+?

LuCaS+ is a multicenter single-cell and single-nucleus RNA-sequencing atlas of non-small-cell lung cancer (NSCLC) that captures the cellular and molecular diversity of this disease across clinical stages and histological subtypes. It comprises more than 2.5 million nuclei from 454 patients with extensive clinical annotation and long-term follow-up, generated from formalin-fixed, paraffin-embedded (FFPE) samples at three German university hospitals. The resource is divided into a discovery cohort (Berlin & Cologne, n = 364) and an independent validation cohort (Munich, n = 90), which have been sampled and analyzed seperately, enabling both biological discovery and external confirmation of results.

By integrating high-resolution single-cell transcriptomic data with detailed clinical and pathological information, LuCaS+ reveals tumor subpopulations with distinct metastatic potential, microenvironmental states associated with outcome, and molecular programs linking oncogenic signaling, epithelial–mesenchymal transition, angiogenesis, and immune evasion.

What you can do

  • Explore patient cohorts: Browse the discovery and validation cohorts, filter by clinical and pathological parameters, and visualize cohort-level distributions.
  • Access raw data: Download individual per-sample CellRanger .h5 outputs or sample metadata.
  • Examine the cell-type hierarchy: Interactively explore the hierarchical taxonomy of epithelial, stromal, and immune cell populations, and inspect differential gene-expression tables for each cell type.
  • Visualize UMAP embeddings: Display all analyzed cells in two-dimensional space and color them by molecular or clinical annotations.
  • Quantify cellular frequencies: Compare the relative abundance of cell types across clinical or molecular groups, or inspect per-sample stacked compositions with metadata overlays.
